LCB22A-exatecan is a preclinical-stage antibody-drug conjugate (ADC) developed by Go Therapeutics in collaboration with LigaChem Biosciences. It consists of the monoclonal antibody L100 (also known as GO-013), which selectively targets cancer-specific O-GalNAc-glycoproteins (Tn-glycosylated proteins) that are aberrantly expressed in approximately 20% of breast cancers and other solid tumors. The antibody is conjugated to the topoisomerase I inhibitor payload, exatecan, via a site-specific, plasma-stable, and tumor-labile β-glucuronidase-cleavable linker utilizing LigaChem's ConjuAll technology. By targeting cancer-restricted glycoepitopes, LCB22A-exatecan minimizes on-target, off-tumor toxicities in healthy tissues. In preclinical models, the ADC has demonstrated potent cytotoxicity and tumor elimination in breast and lung cancer xenografts while maintaining a favorable safety and tolerability profile in non-human primates.
