LCB22A-MMAE (also known as L100-MMAE) is a preclinical-stage antibody-drug conjugate (ADC) being co-developed by Go Therapeutics and LigaChem Biosciences for the treatment of solid tumors, including breast cancer. The ADC consists of the novel monoclonal antibody L100 (GO-013), which selectively targets a cancer-specific O-GalNAc-glycoprotein (Tn-glycosylated protein) expressed in approximately 20% of breast cancers, conjugated to the microtubule-disrupting agent monomethyl auristatin E (MMAE) via a site-specific, plasma-stable, and tumor-labile beta-glucuronidase-cleavable linker. By targeting aberrant O-glycosylation unique to tumor tissues, LCB22A-MMAE minimizes on-target off-tumor toxicities. Preclinical studies have demonstrated potent cytotoxicity and tumor elimination in breast and lung cancer models, with favorable tolerability in non-human primates.
