LCB45A is an antibody-drug conjugate (ADC) targeting aberrant O-glycoproteins, specifically Tn-glycosylated proteins, for the treatment of solid tumors. Developed through a collaboration between GO Therapeutics and LegoChem Biosciences, LCB45A consists of the GO-08 monoclonal antibody conjugated to the microtubule-disrupting payload monomethyl auristatin E (MMAE). The conjugation utilizes LegoChem's proprietary ConjuAll technology, which features a site-specific, beta-glucuronidase-cleavable linker designed to ensure plasma stability and minimize off-target toxicity by releasing the payload primarily within the tumor microenvironment. Preclinical studies have demonstrated significant anti-tumor activity and target-specific cytotoxicity in breast and lung cancer patient-derived xenograft (PDX) models.
