LCB84 is a next-generation antibody-drug conjugate (ADC) that targets the trophoblast cell surface antigen 2 (TROP2), which is highly expressed in various cancer cells. The drug consists of an anti-TROP-2 monoclonal antibody linked to monomethyl auristatin E (MMAE), a potent tubulin inhibitor. Upon binding to TROP-2 on tumor cells and internalization, MMAE is released inside the cell via cleavage by lysosomal enzymes such as beta-glucuronidase. This release disrupts microtubule polymerization, leading to cell cycle arrest and apoptosis of the cancer cell[1][6][7]. Developed using LegoChem Biosciences' proprietary ADC platform technology with a licensed Trop2 antibody from Mediterranea Theranostic Srl, LCB84 has demonstrated differentiated safety and efficacy profiles in preclinical studies across multiple cancers[6]. It is currently being evaluated in Phase 1/2 clinical trials for advanced solid tumors both as a single agent and in combination with an anti-PD-1 antibody[4][5].
