LCB97 is a preclinical-stage antibody-drug conjugate (ADC) developed using LigaChem Biosciences' proprietary ConjuAll ADC platform, targeting the L1 cell adhesion molecule (L1CAM, also known as CD171), which is highly expressed in multiple solid tumors. The drug combines an anti-L1CAM monoclonal antibody (originally developed by Elthera) with a cytotoxic payload, monomethyl auristatin E (MMAE). The mechanism of action involves selective binding to L1CAM-expressing tumor cells, delivery of the MMAE payload, and subsequent inhibition of cell proliferation via tubulin inhibition. LCB97 demonstrates strong anti-tumor activity in preclinical mouse models and is intended as a first-in-class therapy for solid tumors. Ono Pharmaceutical holds exclusive global rights to develop, manufacture, and commercialize LCB97 for solid tumors under a license agreement[1][2][3][4][5][7][8][10].
