LCE62 is a novel, non-RGD-containing linear octapeptide (sequence: MAFKHKAH) that acts as a high-affinity ligand for integrin αvβ3 (KD = 10.8 ± 1.2 nM). Discovered by researchers at the University of Western Ontario using a "beads on a bead" screening approach of a one-bead-one-compound (OBOC) library, LCE62 targets αvβ3 integrin-expressing tumor cells and angiogenic endothelium. Unlike traditional RGD-containing peptides, LCE62 does not disrupt cell adhesion, alter cell morphology, or inhibit angiogenesis, potentially avoiding the risk of promoting tumor invasion and metastasis associated with RGD-mediated anti-angiogenic activity. It is primarily developed as a molecular imaging agent or a vehicle for targeted delivery of diagnostic and therapeutic agents to αvβ3-expressing cancers, such as breast cancer.
