lcLNP anti-CD19 CAR mRNA is an in vivo chimeric antigen receptor (CAR) therapy candidate developed by NanoVation Therapeutics in collaboration with the Fred Hutch Cancer Center. The therapy utilizes NanoVation's proprietary long-circulating lipid nanoparticle (lcLNP) platform to deliver mRNA encoding an anti-CD19 CAR directly to immune effector cells, including T cells and myeloid cells, following intravenous administration. Unlike conventional LNPs that are rapidly cleared by the liver, lcLNPs are engineered for extended circulation and enhanced biodistribution to extrahepatic lymphoid tissues such as the spleen and bone marrow. This approach facilitates the transient in vivo generation of CAR-expressing cells that can effectively deplete B cells, offering a potentially more accessible and scalable alternative to traditional ex vivo CAR-T cell therapies for the treatment of B-cell malignancies and autoimmune diseases like systemic lupus erythematosus (SLE).
