**LCpG** is an experimental lymph-node-targeted cancer-vaccine nanoadjuvant, also termed lipid-glycoadjuvant@AuNPs. It consists of a CpG oligodeoxynucleotide Toll-like receptor 9 agonist covalently modified with lipid-glycopolymers and associated with gold nanoparticles. In mouse melanoma models using ovalbumin as a model antigen, LCpG improved lymph-node retention, promoted antigen-specific CD8-positive Tc1 responses and cytotoxic T-cell infiltration, promoted macrophage M1 polarization, reduced regulatory T-cell differentiation, and generated IFN-gamma-dependent antitumor activity. Its activity was enhanced by co-administration with anti-PD-1 therapy. LCpG has been reported only in preclinical experimental studies and is not an approved medicine.
