Drug intelligence / Profile preview
LD PepGNP-Dengue is a low-dose, gold nanoparticle (GNP)-based, multi-valent synthetic peptide vaccine candidate. Developed by Gylden Pharma Ltd. with support from Emergex Vaccines Holding Limited, it is designed to prime CD8+ T-cell responses against all four serotypes of the dengue virus (DENV). The vaccine utilizes a T-cell priming cocktail of DENV ligandome peptides (2.5 nmol in the low-dose formulation) mounted on gold nanoparticles (14.8 μg gold content). Unlike traditional vaccines that focus on neutralizing antibodies, LD PepGNP-Dengue is engineered to confer protective immunity via cytotoxic T cells without inducing anti-DENV antibodies, thereby minimizing the risk of antibody-dependent enhancement (ADE) and severe dengue. It has been evaluated in a Phase 1 clinical trial (NCT04935801; 'naNO-DENGUE') in healthy adults, where it demonstrated a favorable safety profile and significant increases in dengue-specific CD8+ T-cell memory subsets.
