LD22-4 is a biologic peptide consisting of an 86-amino acid fragment derived from the N-terminal region of the 24-kDa isoform of basic fibroblast growth factor (FGF2). It functions as a potent inhibitor of cell migration by specifically binding to the Neuropilin 1 (NRP1) receptor. Preclinical studies have demonstrated its ability to inhibit the migration of tumor cells, endothelial cells, and fibroblasts, leading to suppressed tumor growth and angiogenesis. Notably, it has shown significant efficacy in orthotopic mouse models of malignant glioma, reducing tumor growth rates by approximately 90%. The peptide's mechanism of action is distinct from the 18-kDa isoform of FGF2, which typically promotes cell proliferation and migration.
