LD78 beta-targeted deoxyribonucleic acid vaccine is an experimental DNA-based cancer vaccine designed for the treatment of B-cell malignancies, including chronic lymphocytic leukemia (CLL) and non-Hodgkin lymphoma (NHL). Developed using the Vaccibody platform, the vaccine encodes a homodimeric fusion protein comprising three functional units: a targeting unit (the human chemokine LD78β, also known as CCL3L1), a dimerization unit (human IgG3 hinge and CH3 domains), and an antigenic unit (tumor-specific single-chain fragment variable, scFv). The mechanism of action involves the targeting of the idiotype antigen to antigen-presenting cells (APCs) via the chemokine receptors CCR1, CCR3, and CCR5. This targeted delivery, often combined with electroporation, enhances the induction of anti-idiotypic antibody and T-cell responses. The vaccine is particularly investigated for its potential as an 'off-the-shelf' therapeutic for patients expressing stereotyped B-cell receptors.
