LDE-paclitaxel is a novel formulation of the chemotherapeutic agent paclitaxel in which the drug is associated with synthetic low-density lipoprotein (LDL)-like lipid core nanoparticles (LDE). This nanotechnology-based delivery system aims to enhance the concentration of paclitaxel in neoplastic cells by mimicking natural LDL particles, potentially increasing antitumor efficacy and reducing systemic toxicity. The mechanism of action remains that of paclitaxel—stabilization of microtubules and inhibition of cell division—but the LDE carrier may improve tumor targeting and tolerability. Clinical studies have evaluated LDE-paclitaxel primarily in advanced pancreatic adenocarcinoma as second- or third-line therapy, showing promising disease control rates and notably low toxicity profiles compared to conventional formulations. Early-phase trials have also explored its safety in cardiovascular disease settings[5][6][8].
