LDN-212320 is a small molecule activator of the glutamate transporter-1 (GLT-1), also known as excitatory amino acid transporter 2 (EAAT2). It functions by increasing the protein expression levels of EAAT2, which is the primary transporter responsible for clearing glutamate from the synaptic cleft in the central nervous system. By enhancing glutamate uptake, LDN-212320 aims to mitigate excitotoxicity and restore glutamatergic homeostasis. Preclinical research has explored its potential in treating various conditions, including chronic pain, cognitive impairment, anxiety, and neurodegenerative diseases. It has demonstrated neuroprotective effects in animal models by upregulating astroglial GLT-1 in key brain regions like the hippocampus and anterior cingulate cortex.
