LDN-5744 is a small molecule inhibitor of Ubiquitin C-terminal hydrolase L1 (UCHL1), a deubiquitinating enzyme that is frequently overexpressed in various malignancies, including high-grade serous ovarian cancer (HGSOC). By targeting UCHL1, LDN-5744 disrupts protein homeostasis and metabolic pathways, leading to reduced cell viability and the sensitization of chemoresistant cancer cells to platinum-based therapies such as carboplatin. Research indicates that UCHL1 inhibition with LDN-5744 results in significant metabolic shifts, including the upregulation of amino acid biosynthesis enzymes and the downregulation of growth-related proteins like CEP55. Furthermore, its efficacy appears to be dependent on the platinum-sensitivity status of the tumor, showing potent activity in chemoresistant models by modulating key signaling pathways such as AKT, STAT3, and ERK.
