Drug intelligence / Profile preview
Lead (212Pb) bamzireotide navoxetan (VMT-alpha-NET) is an investigational, SSTR2-targeted alpha-particle radiopharmaceutical therapy developed by Perspective Therapeutics (originally Viewpoint Molecular Targeting). The drug consists of a somatostatin analog peptide (PSC-PEG2-TOC) that selectively binds to somatostatin receptor subtype 2 (SSTR2), which is highly overexpressed on various neuroendocrine and solid tumors. The peptide is conjugated via a proprietary lead-specific chelator to the alpha-emitting radioisotope lead-212 (212Pb). Upon binding to SSTR2, the radioligand is internalized, delivering high-energy, short-range alpha radiation directly to the tumor cells, causing lethal double-strand DNA breaks while minimizing damage to surrounding healthy tissues. VMT-alpha-NET is currently in Phase 1/2 clinical development for the treatment of SSTR2-expressing tumors, including gastroenteropancreatic neuroendocrine tumors (GEP-NETs), pheochromocytomas, paragangliomas, meningiomas, small cell lung cancer, renal cell carcinoma, and olfactory neuroblastoma.
