LEAF-1702 is a small molecule, liposomal drug developed by L.E.A.F. Pharmaceuticals for the treatment of various cancers. It is a liposomal formulation of gamma-D pentaglutamated pemetrexed-D and acts as an inhibitor of thymidylate synthase (TYMS), disrupting DNA synthesis in rapidly dividing cancer cells. LEAF-1702 belongs to a new generation of onco-immuno antimetabolites designed to disrupt dysregulated serine-glycine 1-carbon metabolism in cancer and the immune system. The drug has reached Phase 1/2 clinical development for indications including head and neck neoplasms, ovarian cancer, and pancreatic cancer[1][2][3][4].
