Overview

Lebrikizumab is a humanized IgG4 monoclonal antibody that targets and neutralizes interleukin-13 (IL-13), a cytokine involved in the pathogenesis of atopic dermatitis and other inflammatory conditions. By binding to IL-13 with high affinity, lebrikizumab inhibits IL-13 signaling through the IL-4 receptor alpha (IL-4Rα)/IL-13 receptor alpha 1 (IL-13Rα1) heterodimer, thereby blocking downstream inflammatory pathways. Unlike some other anti–IL-13 antibodies, it does not prevent IL-13 from binding to its decoy receptor (IL‑13Rα2), allowing for endogenous regulation of IL‑13 clearance. Lebrikizumab is primarily indicated for moderate-to-severe atopic dermatitis in adults and adolescents aged 12 years or older who are candidates for systemic therapy[1][3][5][6]. It is administered by subcutaneous injection.
