Drug intelligence / Profile preview
**ledipasvir + sofosbuvir + vedroprevir** is a triple oral direct-acting antiviral (DAA) combination for the treatment of chronic hepatitis C virus (HCV) genotype 1 infection, especially in patients with compensated cirrhosis who have had prior treatment failures. - **Ledipasvir** is a NS5A inhibitor that blocks the HCV NS5A protein involved in viral replication. - **Sofosbuvir** is a nucleotide analog that inhibits the HCV NS5B RNA-dependent RNA polymerase, preventing viral RNA synthesis. - **Vedroprevir** (GS-9451) is an investigational NS3/4A protease inhibitor targeting viral replication. This combination, studied primarily by Gilead Sciences, demonstrated a 96% sustained virologic response at 12 weeks (SVR12) in a phase 2 trial, without the need for ribavirin. The regimen is developed to maximize antiviral potency and minimize treatment duration in harder-to-treat patients, particularly genotype 1 with cirrhosis.
