Overview

Lefamulin is a semi-synthetic pleuromutilin antibiotic indicated for the treatment of community-acquired bacterial pneumonia (CABP) in adults. It acts by selectively binding to the peptidyl transferase center of the 50S subunit of bacterial ribosomes, thereby inhibiting protein synthesis and preventing peptide bond formation. This unique mechanism disrupts bacterial growth and is effective against a range of respiratory pathogens, including *Streptococcus pneumoniae*, *Staphylococcus aureus* (including methicillin-resistant strains), *Haemophilus influenzae*, *Legionella pneumophila*, *Chlamydophila pneumoniae*, and *Mycoplasma pneumoniae*. Lefamulin is available in both oral and intravenous formulations. Developed and manufactured by Nabriva Therapeutics.
