The legumain DNA vaccine is a preclinical-stage DNA-based cancer vaccine designed to target legumain (asparaginyl endopeptidase or LGMN), a cysteine protease overexpressed on tumor cells, tumor-associated endothelial cells, and tumor-associated macrophages (TAMs). The vaccine typically consists of a DNA plasmid encoding legumain (often fused to polyubiquitin to enhance proteasomal antigen processing). Upon administration, it induces a robust cytotoxic CD8+ T-cell immune response against legumain-expressing cells in the tumor microenvironment, thereby suppressing tumor angiogenesis, growth, and metastasis. Various delivery systems have been investigated, including oral administration via attenuated *Salmonella typhimurium* or alginic acid-coated chitosan nanoparticles (A.C.NPs).
