Leiolizumab is a next‑generation, subcutaneously administered humanized monoclonal antibody being developed by AltruBio as an immune checkpoint enhancer targeting the PSGL‑1 pathway to down‑regulate pathogenic T‑cell effector responses in autoimmune and inflammatory diseases.[3][12][24] Designed as a more potent successor to AltruBio’s first ICE antibody ALTB‑168, leiolizumab selectively agonizes PSGL‑1 on T cells to induce immune tolerance and reduce inflammatory cytokine production, enabling convenient subcutaneous dosing with a favorable nonclinical safety profile.[3][12][24] The lead clinical indication is biologic‑refractory moderately to severely active ulcerative colitis, where leiolizumab is in an ongoing phase 2a open‑label efficacy and safety study following completion of phase 1 evaluation in healthy volunteers.[3][12][15]
