Overview

LEKTI domain 6 is a recombinant protein fragment derived from the lympho-epithelial Kazal-type-related inhibitor (LEKTI), which is a multidomain serine protease inhibitor encoded by the SPINK5 gene. LEKTI domain 6 functions as an efficient serine protease inhibitor, specifically targeting and inhibiting kallikrein-related peptidase 5 (KLK5) and kallikrein-related peptidase 7 (KLK7), as well as trypsin. This inhibition plays a regulatory role in skin desquamation and inflammation. Preclinical studies have demonstrated that topical administration of LEKTI domain 6 reduces inflammatory responses in models of atopic dermatitis and decreases upregulation of pro-inflammatory cytokines such as CXCL-8, CCL20, and IL-6. The mechanism of action involves direct inhibition of serine proteases implicated in skin barrier dysfunctions seen in diseases like Netherton syndrome and atopic dermatitis[2][3][4][5][6]. Development efforts are focused on its potential use as a protein replacement or topical therapy for inflammatory skin conditions.
