Lemzoparlimab is a fully human monoclonal antibody that targets the cell surface antigen CD47. By binding to a unique epitope on CD47 expressed on tumor cells, it blocks the "don't eat me" signal that normally protects cancer cells from being phagocytosed by macrophages. This action enhances macrophage-mediated anti-tumor activity and induces phagocytosis of tumor cells. Lemzoparlimab is designed to minimize binding to normal red blood cells (RBCs), thereby reducing the risk of anemia—a common side effect seen with other CD47-targeting antibodies—while maintaining strong anti-tumor efficacy. The drug has been investigated in combination with agents such as azacitidine and venetoclax for hematologic malignancies like myelodysplastic syndromes (MDS) and acute myeloid leukemia (AML), as well as in combination with pembrolizumab or rituximab for solid tumors and lymphoma[1][3][5][7].
