This is a combination regimen of three drugs: **lenalidomide**, an immunomodulatory agent; **ibrutinib**, a Bruton's tyrosine kinase (BTK) inhibitor; and **rituximab**, a monoclonal anti-CD20 antibody. The triplet is investigated primarily for relapsed/refractory diffuse large B-cell lymphoma (DLBCL), especially the non-germinal center B-cell-like subtype, and mantle cell lymphoma (MCL), as well as central nervous system (CNS) lymphoma and follicular lymphoma. The combination capitalizes on synergy between lenalidomide and ibrutinib, which can result in a pronounced anti-lymphoma effect via immunomodulation, inhibition of NF-κB signaling, reduction of IRF4 expression, and enhanced interferon-β signaling. Rituximab contributes to targeted B-cell depletion by binding CD20 on malignant B cells, augmenting the immune response. Clinical trials indicate high response rates, manageable toxicity, and potential for durable remissions, particularly in previously treated or high-risk populations[1][2][3][9].
