Overview

Lenalidomide + vorinostat is an investigational combination therapy primarily studied for the treatment of relapsed or refractory multiple myeloma. Lenalidomide is an immunomodulatory agent and thalidomide derivative that works by altering cytokine production, regulating T cell co-stimulation, enhancing NK cell-mediated cytotoxicity, and promoting degradation of key B-cell transcription factors through cereblon binding. Vorinostat is a small molecule histone deacetylase (HDAC) inhibitor that induces cell cycle arrest and apoptosis in malignant cells by altering gene expression via epigenetic modulation. Preclinical studies suggest synergy between these agents, with clinical trials showing moderate response rates in patients with relapsed/refractory multiple myeloma[1][3][5][6]. Both drugs are orally administered small molecules.
