Overview

lenalidomide + CHOP is a combination chemotherapy regimen consisting of the immunomodulatory drug lenalidomide and the standard CHOP backbone (cyclophosphamide, doxorubicin, vincristine, and prednisone). Lenalidomide is a small molecule that binds to the cereblon E3 ubiquitin ligase complex, leading to the degradation of transcription factors like Ikaros (IKZF1) and Aiolos (IKZF3), which results in antiproliferative and immunomodulatory effects. CHOP is a multi-agent chemotherapy regimen that targets DNA replication and microtubule assembly. This specific combination was evaluated by The Lymphoma Academic Research Organisation (LYSARC) as a first-line treatment for elderly patients (aged 60-80) with angioimmunoblastic T-cell lymphoma (AITL). In clinical trials, the regimen aimed to improve complete metabolic response rates, although results indicated it did not significantly outperform historical controls in this specific population.
