Lenti-IiE1 is an investigational lentiviral vector-based therapeutic vaccine developed by Theravectys for the treatment of diseases caused by Human Papillomavirus type 16 (HPV16), including cervical intraepithelial neoplasia (CIN) and oropharyngeal cancers. The vaccine utilizes a non-replicative, self-inactivating lentiviral vector to deliver the genetic sequence of the HPV16 E1 protein. A distinguishing feature of this construct is the fusion of the E1 antigen to the MHC class II-associated invariant chain (Ii/CD74). This fusion directs the antigen to the endosomal compartment, enhancing its processing and presentation on MHC class II molecules. This mechanism is designed to significantly boost CD4+ T-cell help and the subsequent activation of robust, long-lasting cytotoxic CD8+ T-cell responses. The ultimate therapeutic goal is the targeted elimination of HPV16-infected cells and the prevention of disease progression to invasive malignancy.
