Lenti-Lα5 shRNA is a lentiviral-based RNA interference (RNAi) agent designed to silence the expression of the laminin α5 chain (LAMA5). LAMA5 is a critical component of laminin-511 (α5β1γ1), a major basement membrane protein that is frequently upregulated in malignant tumors, including melanoma. By delivering specific short hairpin RNA (shRNA) via a lentiviral vector, this agent facilitates the degradation of LAMA5 mRNA, thereby preventing the synthesis and secretion of laminin-511. Preclinical research conducted at the University of Miami has demonstrated that knockdown of LAMA5 using Lenti-Lα5 shRNA significantly inhibits melanoma cell proliferation, attachment, migration, and invasion in vitro. Furthermore, in vivo studies in nude mice models have shown that blocking laminin-511 expression markedly reduces tumor growth, suggesting that LAMA5 is a viable therapeutic target for melanoma.
