lentiviral hCD68-mCherry engineered hematopoietic stem cells is a preclinical cell and gene therapy platform developed by researchers at Northwestern University. It consists of autologous hematopoietic stem cells (HSCs) transduced with a lentiviral vector containing the human CD68 (hCD68) promoter, which drives the expression of a transgene—in this specific research construct, the fluorescent reporter mCherry. The platform is designed for glioma immunotherapy, leveraging the natural ability of HSC-derived myeloid cells to infiltrate the tumor microenvironment and differentiate into tumor-associated macrophages (TAMs). The hCD68 promoter ensures that transgene expression is restricted to the macrophage lineage and is significantly upregulated within the immunosuppressive tumor microenvironment, providing a modular system for targeted delivery of therapeutic agents to glioblastomas and other macrophage-rich malignancies.
