Lenvatinib + rAd-p53 is a combination therapy consisting of lenvatinib, a small molecule multi-kinase inhibitor, and rAd-p53 (recombinant adenoviral p53 gene therapy). Lenvatinib targets multiple receptor tyrosine kinases involved in tumor angiogenesis and proliferation, including vascular endothelial growth factor receptors (VEGFR), fibroblast growth factor receptors (FGFR), and platelet-derived growth factor receptor beta (PDGFR-β). rAd-p53 delivers the wild-type p53 gene to tumor cells via an adenoviral vector, aiming to restore normal p53 function and induce apoptosis in cancer cells with dysfunctional or mutated p53. The combination has been studied for its synergistic anti-tumor effects in cancers such as non-small cell lung cancer (NSCLC) and renal cell carcinoma (RCC), showing enhanced inhibition of tumor growth, migration, invasion, and increased apoptosis compared to either agent alone. Common adverse events include hypertension, diarrhea, nausea, proteinuria, body weight loss for the combination; fever is more common with gene therapy components[1][2][3].
