Overview

Lenvatinib + tislelizumab + FOLFOX is a combination therapeutic regimen being evaluated for the treatment of advanced hepatocellular carcinoma (HCC). This regimen integrates three distinct therapeutic approaches: lenvatinib, a multi-kinase inhibitor that targets vascular endothelial growth factor receptors (VEGFR 1-3) and fibroblast growth factor receptors (FGFR 1-4) to inhibit angiogenesis and tumor growth; tislelizumab, a humanized IgG4 monoclonal antibody that inhibits the programmed cell death protein 1 (PD-1) receptor to enhance anti-tumor immune responses; and FOLFOX, a standard chemotherapy regimen comprising oxaliplatin, 5-fluorouracil (5-FU), and leucovorin (folinic acid). The combination aims to achieve synergistic anti-tumor activity by simultaneously targeting tumor vascularization, overcoming immune evasion, and inducing cytotoxic cell death. Clinical investigation, primarily led by The Third Affiliated Hospital of Sun Yat-sen University, focuses on its efficacy as a first-line treatment for patients with advanced HCC.
