Lenzilumab is a humanized monoclonal antibody (IgG1 kappa) that targets and neutralizes granulocyte-macrophage colony-stimulating factor (GM-CSF), also known as colony stimulating factor 2 (CSF2). Developed using Humaneered technology to optimize its properties, lenzilumab binds GM-CSF with high affinity and specificity, preventing its signaling through the GM-CSF receptor. This mechanism interrupts the inflammatory cascade initiated by GM-CSF, which plays a key role in cytokine release syndrome (CRS), particularly in patients undergoing chimeric antigen receptor T cell (CAR-T) therapy and those hospitalized with severe COVID-19 pneumonia. Lenzilumab is being developed primarily for chronic myelomonocytic leukemia (CMML), where it is studied both as monotherapy and in combination with azacitidine. It has also been investigated for prevention or treatment of CRS associated with CAR-T therapy, acute graft versus host disease, juvenile myelomonocytic leukemia, myelodysplastic syndromes, myeloproliferative neoplasms, acute myeloid leukemia, and severe COVID-19[1][5][6][7][8].
