Overview

LEO 134310 is a selective, non-steroidal glucocorticoid receptor (GR) agonist developed by LEO Pharma for topical use in the treatment of inflammatory skin diseases such as plaque psoriasis and atopic dermatitis. Unlike traditional steroidal glucocorticoids, it is rapidly deactivated by enzymatic hydrolysis in the blood, resulting in low systemic exposure and a higher therapeutic index. This property aims to reduce local and systemic side effects commonly associated with conventional topical steroids. Preclinical studies have shown that topical application of LEO 134310 does not cause significant epidermal thinning or skin atrophy compared to standard treatments like betamethasone valerate or clobetasol propionate. The drug has demonstrated high affinity for the glucocorticoid receptor (EC50 of 14 nM) and selectivity over other nuclear receptors[1][2][3][4][5][6][7].
