Overview

LEO 39652 is a "dual-soft" small molecule phosphodiesterase 4 (PDE4) inhibitor developed by LEO Pharma for topical treatment of atopic dermatitis. The compound was designed with ester functionalities to enable rapid inactivation in blood and liver, thereby reducing systemic side effects while maintaining stability and efficacy in the skin. Preclinical studies demonstrated high local exposure with minimal systemic absorption, supporting its use as a topical therapy. Clinical development reached phase I trials for atopic dermatitis but has not advanced further as of the latest updates[1][2][3][8].
