Leptin + vasodilators refers to a hypothetical or experimental combination of the hormone leptin with one or more drugs that act as vasodilators. Leptin is an adipocyte-derived hormone primarily involved in regulating appetite and energy expenditure, but it also has complex cardiovascular effects, including increasing sympathetic nerve activity and potentially influencing vascular tone. Vasodilators are a broad class of pharmaceutical agents that relax blood vessels, thereby lowering blood pressure and improving blood flow; they are commonly used in conditions such as hypertension, heart failure, and angina. Mechanistically, some studies suggest that leptin can induce endothelium-dependent vasorelaxation via nitric oxide (NO) and endothelium-derived hyperpolarizing factor (EDHF) pathways in isolated vessel preparations[5][7]. However, other research indicates that at physiologically relevant concentrations in vivo, leptin does not exert significant direct vasodilator effects; its predominant action is to increase sympathetic nerve activity which may counteract any potential peripheral vasodilation[1][6][9]. The combination of exogenous leptin with pharmacological vasodilators has not been established as a clinical therapy nor widely studied as a fixed drug product.
