Leptoprol

Leptoprol is a sustained-release implant formulation containing 5 mg of leuprorelin acetate, a synthetic nonapeptide analog of naturally occurring gonadotropin-releasing hormone (GnRH). Developed and marketed by Recordati, it is primarily indicated for the palliative treatment of advanced, hormone-dependent prostate cancer. As a GnRH receptor agonist, leuprorelin initially stimulates the release of luteinizing hormone (LH) and follicle-stimulating hormone (FSH) from the pituitary gland. However, continuous administration leads to the downregulation and desensitization of pituitary GnRH receptors, resulting in a significant suppression of gonadotropin secretion. This suppression reduces serum testosterone to castrate levels, thereby inhibiting the growth of hormone-sensitive prostate cancer cells. The 5 mg implant is designed to provide therapeutic levels of the drug over a three-month period.
