Overview

RAY1216 (generic name: leritrelvir) is a small molecule oral inhibitor of the SARS-CoV-2 3C-like protease (also known as main protease or Mpro), developed for the treatment of COVID-19. It covalently binds to the catalytic cysteine residue of the viral protease, thereby blocking viral replication. Ritonavir is co-administered as a pharmacokinetic booster; it inhibits CYP3A-mediated metabolism of RAY1216, increasing its plasma exposure. Clinical studies have shown that both RAY1216 monotherapy and combination with ritonavir significantly reduce viral load and duration of COVID-19 symptoms compared to placebo, with a favorable safety profile. However, due to improved pharmacokinetics and efficacy as monotherapy, use without ritonavir is also being pursued[1][3][4][6].
