Drug intelligence / Profile preview
leronlimab + TAS-102 + bevacizumab is an investigational combination regimen being evaluated for the treatment of previously treated, refractory microsatellite stable (MSS) metastatic colorectal cancer (mCRC). The regimen combines leronlimab (formerly PRO 140), a humanized monoclonal antibody targeting the CCR5 chemokine receptor, with the standard-of-care chemotherapy backbone consisting of TAS-102 (trifluridine/tipiracil) and the anti-VEGF-A monoclonal antibody bevacizumab. CCR5 is implicated in the recruitment of immunosuppressive myeloid cells and regulatory T cells to the tumor microenvironment, promoting immune evasion. By blocking CCR5, leronlimab is intended to modulate the tumor microenvironment and enhance the anti-tumor activity of the cytotoxic and anti-angiogenic therapies. The combination is currently being evaluated in the Phase 2 CLOVER trial (NCT06699836).
