This is a **four-drug anti-cancer combination** composed of leronlimab, trifluridine, tipiracil, and bevacizumab. - **Trifluridine** is a thymidine-based nucleoside analog that inhibits thymidylate synthase and incorporates into DNA, leading to DNA damage and cell death. - **Tipiracil** is a thymidine phosphorylase inhibitor that prevents enzymatic breakdown of trifluridine, increasing its efficacy. - **Bevacizumab** is a monoclonal antibody that targets vascular endothelial growth factor A (VEGF-A), inhibiting angiogenesis required for tumor growth. - **Leronlimab** is a humanized monoclonal antibody targeting the CCR5 receptor, being investigated for various cancer and infectious disease indications. Trifluridine + tipiracil (Lonsurf) in combination with bevacizumab (Avastin) is FDA approved for treatment-refractory metastatic colorectal cancer. Leronlimab is not an approved part of this combination for colorectal cancer and is primarily in development.
