Overview

Lesofavumab (MHAB5553A) is a human monoclonal immunoglobulin G1 (IgG1) antibody developed as a novel therapeutic for the treatment of severe influenza B virus infection. It binds specifically and with high affinity to a highly conserved epitope on the esterase domain of the hemagglutinin (HA) globular head of influenza B virus, thereby preventing HA-mediated membrane fusion in the endosome. Lesofavumab also binds to HA on the surface of infected cells and, through its Fc domain, induces antibody-dependent cell-mediated cytotoxicity (ADCC) by engaging Fcγ receptor-bearing immune cells. The drug has demonstrated linear serum pharmacokinetics and was generally well tolerated in healthy volunteers up to single intravenous doses of 10,800 mg in phase 1 studies[1][2][3].
