Lesogaberan + proton pump inhibitor is an investigational combination therapy used in clinical trials for patients with gastroesophageal reflux disease (GERD) who have an inadequate response to proton pump inhibitor (PPI) monotherapy. Lesogaberan is a novel, peripherally selective γ-aminobutyric acid type B (GABA-B) receptor agonist that reduces transient lower esophageal sphincter relaxations (TLESRs), a major mechanism of reflux. PPIs suppress gastric acid production. The combination aims to address persistent reflux symptoms by targeting both acid suppression (PPI) and TLESR reduction (lesogaberan). Clinical trials showed statistically significant but modest improvements in symptom control when lesogaberan was used as an add-on to PPI therapy in GERD patients.
