Overview

let-7b mimic is a **synthetic microRNA (miRNA) therapeutic** designed to replicate the function of endogenous let-7b microRNA. The let-7 family is one of the first identified and most evolutionarily conserved miRNA families across species. let-7b mimic functions by **post-transcriptionally regulating gene expression** through binding to complementary sequences in the 3' untranslated regions (3' UTRs) of target mRNAs, leading to their degradation or translational repression. The therapeutic mechanism involves targeting multiple oncogenes and regulatory proteins. Key molecular targets include **TLX (nuclear receptor)**, **cyclin D1 (cell cycle regulator)**, **HMGA2 (high mobility group protein)**, **HK2 (hexokinase 2)**, and immune checkpoint molecules **PD-1** and **PD-L1**. By suppressing these targets, let-7b mimic inhibits cancer cell proliferation, promotes differentiation, reduces glycolytic metabolism in cancer cells, and enhances anti-tumor immune responses. Various formulations have been developed to improve delivery and stability, including **hydrophobically modified versions (hmiRNA)** conjugated to lipids such as eicosapentaenoic acid (EPA), docosanoic acid (DCA), or cholesterol. These modifications enhance cellular uptake, serum stability, and biodistribution to target tissues including lungs and tumors. An **aerosolized pulmonary delivery formulation** has been specifically developed for lung cancer applications.
