Leteprinim potassium is a synthetic small molecule and hypoxanthine derivative with neuroprotective and nootropic properties. It acts as a nerve growth factor modulator, stimulating the release of nerve growth factors and enhancing neuronal survival in the brain. Leteprinim has demonstrated efficacy in preclinical models by promoting neurite outgrowth, increasing synaptophysin production, improving working memory in aged mice, reducing locomotor impairment after spinal cord injury, and protecting against hypoxia-induced neuronal loss. Its mechanism involves modulation of neurotrophic pathways relevant to neurodegenerative diseases such as Alzheimer's disease, Parkinson's disease, stroke, spinal cord injury, and potentially depression or neonatal hypoxic ischemic encephalopathy. The drug was developed by Spectrum Pharmaceuticals but development for major indications has been discontinued[1][2][3][4][5].
