Letrazuril

Letrazuril is a small molecule investigational drug, structurally related to diclazuril, and is the p-fluoro analogue of diclazuril. It has been studied primarily for the treatment of cryptosporidiosis in severely immunocompromised patients, such as those with AIDS, and has also been investigated for HIV infections. In clinical trials, it showed modest and often short-lived efficacy in reducing symptoms of refractory cryptosporidial diarrhea in AIDS patients. The mechanism of action is presumed to be similar to other triazine derivatives like toltrazuril—primarily affecting the mitochondrial respiratory chain and enzymes involved in pyrimidine synthesis within protozoan parasites. Letrazuril was initially developed by Janssen Global Services but did not progress beyond early-phase clinical trials due to limited efficacy and side effects such as rash[1][2][3][5].
