Letrozole + goserelin + leuprolide is a combination endocrine therapy regimen used in the treatment of estrogen receptor (ER)-positive breast cancer, particularly in premenopausal women. Letrozole is an aromatase inhibitor that works by blocking the enzyme aromatase, thereby preventing the conversion of androgens to estrogens and reducing circulating estradiol levels. Goserelin and leuprolide are both synthetic luteinizing hormone-releasing hormone (LHRH) agonists that suppress ovarian function by inhibiting pituitary gonadotropin secretion, effectively inducing a medically menopausal state in premenopausal women. In this regimen, goserelin or leuprolide is used interchangeably for ovarian function suppression (OFS), after which letrozole is administered to further suppress estrogen. The combination is being evaluated in a Phase II clinical trial context (sponsored by The Second Hospital of Anhui Medical University) as neoadjuvant endocrine therapy combined with chemotherapy in patients with ER-positive, HER2-negative, Ki67 > 20% stage I–III invasive breast cancer.
