Letrozole + leuprolide

Letrozole + leuprolide is a combination therapy used primarily in the treatment of hormone receptor-positive (HR+), HER2-negative breast cancer in premenopausal and perimenopausal women. Letrozole is an aromatase inhibitor that reduces estrogen production by inhibiting the aromatase enzyme responsible for converting androgens to estrogens. Leuprolide is a gonadotropin-releasing hormone (GnRH) agonist that initially stimulates but then suppresses pituitary release of luteinizing hormone (LH) and follicle-stimulating hormone (FSH), leading to decreased ovarian estrogen production. This combination achieves profound estrogen suppression by targeting both peripheral conversion and ovarian synthesis of estrogens[1][3][4]. The regimen is also studied or used off-label for other indications such as endometriosis-related infertility.
