Letrozole + pelareorep is a combination therapy being investigated primarily for breast cancer treatment. Pelareorep is an intravenously delivered immuno-oncolytic reovirus that selectively replicates in tumor cells, promoting immune system activation against cancer. When combined with letrozole (an aromatase inhibitor), this therapy aims to enhance anti-tumor immune responses. The combination works by pelareorep infecting and replicating within tumor cells, leading to increased tumor inflammation, T cell recruitment, and PD-L1 expression, which may improve outcomes when combined with checkpoint inhibitors.
