letrozole-matched placebo

Letrozole-matched placebo is an inactive pharmacological control formulated as an oral tablet to match the physical appearance and administration schedule of letrozole 2.5 mg. It was specifically utilized by Hoffmann-La Roche (Genentech) in the Phase III persevERA clinical trial (NCT04546009) to maintain the double-blind design. In this study, patients in the experimental arm received giredestrant and palbociclib combined with the letrozole-matched placebo, while the control arm received active letrozole and palbociclib. The trial evaluated these regimens in patients with estrogen receptor (ER)-positive, human epidermal growth factor receptor 2 (HER2)-negative locally advanced or metastatic breast cancer. Although the study did not meet its primary endpoint of statistically significant improvement in progression-free survival, a numerical improvement was observed in the giredestrant-containing arm.
