letrozole placebo

Letrozole placebo is an inactive oral formulation designed to physically and visually mimic the aromatase inhibitor letrozole. It is specifically utilized as a blinding control in AstraZeneca's SERENA-6 Phase III clinical trial (NCT04964934). This trial evaluates the efficacy of switching to camizestrant (AZD9833), a next-generation oral selective estrogen receptor degrader (SERD), in combination with a CDK4/6 inhibitor versus continuing an aromatase inhibitor (letrozole or anastrozole) plus a CDK4/6 inhibitor in patients with HR+/HER2- metastatic breast cancer who have detectable ESR1 mutations. In the experimental arm, patients receive active camizestrant along with this letrozole placebo to ensure the study remains double-blinded against the active control arm.
