Overview

LEU011 is an autologous (self-derived) chimeric antigen receptor T-cell (CAR-T) therapy developed by Leucid Bio for the treatment of solid tumors. It is engineered from a patient’s own T lymphocytes, which are genetically modified to express a lateral CAR targeting natural killer group 2 member D ligand (NKG2DL). The NKG2D receptor is an activating immune receptor that recognizes one or more of eight human NKG2D ligands commonly expressed on transformed, infected, or damaged cells—these ligands are present in over 80% of human tumors. Upon recognition of these ligands on tumor cells, the modified T-cells can trigger cell death and stimulate an antitumor immune response. LEU011 uses a novel lateral CAR platform designed to improve tumor targeting and selectivity compared to traditional linear CAR constructs. The drug is currently being evaluated in Phase 1/2 clinical trials for relapsed or refractory solid tumors[1][3][4][5][6].
